Title:A study of the phylogenetics, nutrition and one-carbon metabolism of Syntrophococcus sucromutans

Abstract:Syntrophococcus sucromutans is phylogenetically included in the Gram-positive phylum of the Eubacteria, by virtues of 16S ribosomal RNA sequence similarity analysis. It is part of a well defined genealogic entity described as the Butyrivibrio subdivision which, like the Sporomusa subdivision contains both ultrastructurally Gram-positive and Gram-negative bacteria.
The development of a growth medium devoid of rumen fluid has led to the description of a unique lipid requirement satisfied by octadecenoic acids exclusively, when fatty acids are used as sole lipid supplement, or by various phospholipids and triglycerides. The fatty acid and aldehyde composition of the cells was observed to be highly influenced by that of the lipid supplement and incorporation of (1-$\sp{14}$C) oleic acid indicated a predominant labeling of the membranes, most of the label being recovered during extraction of the fatty acid and aldehyde fractions.
S. sucromutans, isolated as the predominant ruminal species capable of degrading methoxy substituents of monoaromatic compounds derived from lignin, was shown to incorporate label from chemically synthesized O- (methyl-$\sp{14}$C) vanillate exclusively in the methyl group of acetate. Further studies on the incorporation of label from CO$\sb2$, pyruvate and formate, and on the enzymatic components using resting cells and cell extracts finally indicated that S. sucromutans is likely to depend on the Acetyl-CoA pathway for the synthesis of acetate from one carbon compounds and sugars or pyruvate.
