Abstract:The enhancement of fluorescence output is of great significance in the fields of photonics and flurorecence biosensors. We developed an instrument targeting the enhancement of fluorophores that had been spotted on photonic crystal devices. We also sought to investigate the optimization of photonic crystal devices for fluorescence enhancement. Rigorous coupled-wave analysis was used to determine optimal dimensions for the photonic crystal structures, and devices were fabricated using a polymer-based nanoreplica molding approach. The prototype instrument was used to successfully perform enhanced-fluorescence detection on these devices using arrays of spots of strepavidin-Cy-5. Finally, we demonstrate the use of our instrument in performing a simple DNA-microarray analysis utilizing enhanced fluorescence and label-free imaging techniques, for improved quality control and sensitivity.
