Title:Photosynthetic Characteristics of Two Photoautotrophic Cell Suspensions of Soybean

Abstract:A soybean suspension culture (SB-P), grown photoautotrophically in 5% CO$\sb2$, has unique characteristics not found in mature leaf cells. A new photoautotrophic soybean suspension culture (PI-P) was initiated and characterized for the purpose of comparing the physiology with that of SB-P.
Traits shared by SB-P and PI-P are an inability to grow at air levels of CO$\sb2$, active cell division, high dark respiration, low ribulose-1,5-bisphosphate carboxylase (RuBPcase), a low ratio of RuBPcase/phosphoenolpyruvate carboxylase (PEPcase), an elevated CO$\sb2$ compensation concentration in culture, low photosynthetic capacity, and RuBPcase deactivation at air CO$\sb2$ levels.
It was concluded that many of the characteristics of photoautotrophic soybean cultures are caused by the conditions employed in culture, and are not due to the specific genotype of soybean cultured or to chance occurrence during initiation. The cultures share several traits with developing leaf cells (active cell division, low photosynthetic capacity, elevated respiration, a low ratio of RuBPcase(initial)/PEPcase which would be expected since the cultured cells are growing. This indicates that the photoautotrophic cell physiology is more comparable to that of developing leaf cells, rather than to that of mature leaf cells.
The specific activity of RuBPcase, measured immunologically, varies with time in culture, and this is not due to the presence of the RuBPcase inhibitor which is found in soybean plants in the dark. However if the photoautotrophic cells are given a dark period the inhibitor is present. If growth rates are slowed, the cells photosynthetic capacity increases due to increased RuBPcase (in vivo) activity.
