Abstract:The dissertation is concerned with biophysical applications of Forster resonance energy transfer (FRET) and development and application of fluorescence lifetime-resolved imaging microscopy (FLIM). It includes work to extend the FRET and FLIM methods for studying microsecond reaction kinetics. The common thread and unifying theme of the dissertation is the development and application of fluorescence spectroscopy and microscopy (in particular, FRET and FLIM) to biology and medicine. Several separate research topics are presented, including: investigation of the structural properties of a metal-DNA complex (M-DNA); development and characterization of a genetically encoded biosensor for monitoring intracellular redox potentials; image analysis for FLIM; and, a microfluidic mixing device for accessing microsecond reaction kinetics.
