Title:Selection and Characterization of a Steroid Receptor DNA Binding Domain-Based Repressor of Estrogen Response Element Containing Promoters

Abstract:To generate a repressor of the ERE-containing genes, we dimerized one of the up-binding mutants, DBD5, with a flexible linker and fused it to a strong KRAB repressor domain. The dimerized FLAGDBD5-KRAB (FdDBD5K) is a much more potent repressor than the monomer FLAGDBD5-KRAB (FDBD5K). The FdDBD5K elicited significant repression of both constitutive and estrogen receptor induced transcription on a cERE containing promoter and on a natural pS2 promoter. The FdDBD5K chimera should be a useful new tool for controlling the transcription of ERE-containing promoters.
