Title:Characterization of a Pathogenicity Gene in Pseudomonas Syringae Pv. Tabaci Strain PTBR2.024

Abstract:In planta growth curves showed that PTBR4.000 could not reach the same population level in tobacco or green bean leaves as PTBR4.000. A 4.2 kb Hin dIII-Eco RI fragment cloned downstream of a vector promoter was able to restore pathogenicity to PTBR4.000 in intact tobacco leaves. ORF1 and ORF2 were found not to be required for pathogenicity of tobacco since a marker exchange mutation in these ORFs remained pathogenic. The combined data indicate that ORF3 probably was mutated in a polar manner by Tn5 resulting in the loss of tobacco pathogenicity in PTBR4.000. Based on the similarity of ORF3 to an aspartate aminotransferase and the observed phenotypic differences it was proposed that PTBR4.000 is defective in nitrogen metabolism.
